The purpose of this work is pursue the fundamental question of how mechano sensitive neurons sense external forces and convert this physical stimulus into a bio- chemical one. In this process, cell mechanical properties play an important role. This thesis will attempt to incorpórate two different experimental techniques to investigate neuron mechanical properties: first, master basic C. elegans culturing techniques and embryonic cell cultures to isolate mechano sensitive neurons responsible for the sense of touch. Second, establish a forcé stimulation assay exploiting the superior forcé resolution of a optical tweezer to apply minute forcé onto these cells. In summary, the forcé range and resolution, size scale, and position resolutions required to probe mechano sensitive neurons make optical tweezers an attractive option for these experiments. The first goal of the thesis aimed to characterize the mechanical properties of mechano sensitive neurons is isolate and characterize it. Identified neurons responsible for the sense of touchin C. elegans, will proceed to perform the process of isolation. For the second research goal of the thesis, we need adapt optical tweezer assay for force measurements on living neurons. The optical tweezer setup is mounted on a uorescence microscope, with which will monitor the activation of the transgenic mechanosensors by observing their calcium levels using genetically encoded neuronal activity indicators during the force application. This is a proposed method to inves tigate the force neccesary to activate mechanosensitive channels. Finally, like third goal we characterize the neuron mechanical properties via mem brane tethering experiments. Measuring tether extension force on membrane neuron is possible estimate mechanical properties.
